The role of the ubiquitin-protein ligase Nedd4-2 on the electrogenic Na+/HCO3 -cotransporter, NBCe1-B and Pendrin in the thyroid

Supported project of Alain Boom, Renaud Beauwens & Antonina Khoruzenko (IRIBHM-ULB) : The balance between insertion and reuptake of a defined membrane protein, determining its number in the cell membrane. The Na-HCO3 co-transporter NBCe1, located in the basolateral membrane of the thyroid follicular cells of the thyrocyte, is a protein with a short half-life. Its half-life is significantly prolonged upon exposure to the hormone TSH by a mechanism yet to be determined. The present study aims to investigate the expression of Nedd4-2 and its possible interaction with NBCe1 leading to its ubiquitination and subsequent internalization and degradation. The second objective will be to test whether pendrin is also the site of interaction withNedd4-2, leading to its ubiquitination and subsequent degradation.
